The Will is from the Prerogative Court of Canterbury Wills 1384-1848.
I agree the Prudence named in the Will is the daughter.
The query re Prudence Heartland the wife is that he mentions being separated from his wife in the second paragraph but according to information from Ancestry she died in 1806. Humphrey didn't die until 1827 and the children's marriages also post date 1806.

The Thomas Manger I have in the tree is born 1797, died 1860 parents William Manger and Pamela Barrows.
I have no record of Mary Brettell and struggling to find information. She is however mentioned in another family tree but they discount Mary Raybould.

Looks like the name, Ann was used instead of her birth name of Nancy on 1841 Census
